Research
Protein quality control is essential for the proper maintenance of all living cells. It ensures, not only the correct folding of newly synthesised proteins, the refolding or degradation of misfolded and aggregated proteins, but also controls the degradation of regulatory proteins. Collectively, these functions are performed by molecular chaperones and proteases. In our laboratory we are primarily interested in proteases from the bacterial cytosol and the mitochondrial matrix. Our current research concentrates on defining the molecular basis of substrate selectivity for several different ATP-dependent proteases. A major focus of the work is directed towards the mechanistic analysis of novel AAA+ adaptor proteins such as ClpS, which we recently discovered and how these protein co-factors modulate AAA+ substrate specificity. A detailed analysis of protease substrate specificity is central to our understanding of how chaperones and protease compete for different substrates in vivo.
