We are interested on the kallikreins, a group of 15 serine proteases whose genes are localized on human chromosome 19q13.4. All these proteases have significant homologies at the DNA and protein level and exhibit either trypsin or chymotrypsin-like activity. Our laboratory has cloned a number of these proteases and established the genomic organization of the human locus.
One member of this family is prostate-specific antigen (PSA), a valuable biomarker for prostatic carcinoma. We have developed recombinant proteins, antibodies and immunological assays for most of the newly discovered kallikreins. We have shown that a number of them are valuable biomarkers for ovarian, prostate and breast carcinomas. We are currently investigating the enzymatic activity of these proteases, the substrates that they cleave and their participation in physiological and pathological processes, including cancer invasion and metastasis. We are further interested to examine if any of these proteases are targets for cancer or other disease therapy.
